Effects of Hypotensive and Heart Rate of Cyclobuxine in Rats



Abstract
The study was undertaken to search for a new hypotensive agent in natural crude plants. Extracts of Buxus microphylla var. korean¢¥a Nakai have been used as folk remedies of several diseases, including malaria and venereal disease. But any study on the pharmacological action of this plant has not yet been carried out and its active ingredients have not been identified. On the previou studies, buxuletin(nonalkaloid) was isolated from Buxus microphylla var. koreana and its pharmacological action reported to the diuretic effect in rabbits.
In the present study, we have isolated alkaloid from the acetone-insoluble fraction of the strong bases of this plant. This alkaloid(CzsH42ONz) was identified as a steroidal alkaloid contained a cyclopropane ring by physical and chemical methods. We named this alkaloid cyclobuxine. Therefore, we intended to determine the pharmacological action of cyclobuxine, on the blood pressure and the heart rate, in anesthetized rats several drugs were pretreated prior to cyclobuxine to clarify the mechanism of the hypotensive action.
The observed results are as follows
1) Cyclobuxine(lmg/kg), when intravenously administered to rats, significantly lowered the blood pressure and decreased the heart rate.
2) The hypotensive response of cyclobuxine(lmg/kg) was slightly antagonized by pretreatment with phentolamine(2mg/kg) and, in the pretreated groups with propranolol(lmg/kg), was similar to that of control groups.
